A Study to Evaluate ALN-6400 in Healthy Volunteers and Patients With Hereditary Hemorrhagic Telangiectasia (HHT)

Objective

To assess the safety, tolerability, pharmacokinetics (PK), and pharmacodynamics (PD) of the investigational drug ALN-6400 in both healthy volunteers and adult patients diagnosed with Hereditary Hemorrhagic Telangiectasia (HHT).

Methods

The study will employ a two-part design: initially administering single ascending doses of ALN-6400 to healthy volunteers followed by multiple-dose evaluations in patients with HHT. Safety and tolerability will be systematically monitored alongside pharmacokinetic and pharmacodynamic assessments.

Results

Initial findings will provide data on the drug's safety profile and tolerability, while efficacy in HHT patients will be determined through PD responses and symptom assessments related to arteriovenous malformations (AVMs) commonly seen in HHT.

Limitations

The study's limitations may include a small sample size, reliance on short-term observation for efficacy outcomes, and variability in patient responses due to the heterogeneous nature of HHT's clinical presentation.
